About CABENUVA

Overview

The supplied FDA label covers VOCABRIA (cabotegravir) tablets, not CABENUVA. VOCABRIA is used with rilpivirine for short-term HIV-1 treatment in adults and adolescents aged 12 or older who weigh at least 35 kg, have well-controlled HIV on stable treatment, and have no history of treatment failure or known or suspected resistance to either medicine. It is also used for short-term prevention of sexually acquired HIV-1 in at-risk adults and adolescents weighing at least 35 kg, after a negative HIV-1 test. Cabotegravir works by blocking a viral enzyme called integrase.

What it treats

• Cabotegravir tablets (VOCABRIA), combined with rilpivirine, are used for short-term treatment of HIV-1 in adults and adolescents aged 12 years or older weighing at least 35 kg, with a suppressed viral load on stable treatment, no history of treatment failure, and no known or suspected resistance to either medicine. • VOCABRIA is also used for short-term pre-exposure prophylaxis (PrEP) to reduce the risk of sexually acquired HIV-1 in at-risk adults and adolescents weighing at least 35 kg. A negative HIV-1 test is required before starting.

Side effects

For cabotegravir tablets, common side effects reported in people with HIV-1 include tiredness, headache, diarrhoea, nausea, dizziness, unusual dreams, anxiety, difficulty sleeping, abdominal discomfort, bloating and weakness. In people without HIV-1, common side effects include headache, diarrhoea, nausea, dizziness, upper respiratory tract infection, sleepiness, tiredness, unusual dreams and abdominal pain. Serious allergic reactions, including severe skin reactions called Stevens–Johnson syndrome and toxic epidermal necrolysis, have been reported with cabotegravir. Stop taking the tablets and seek immediate medical attention if signs of an allergic reaction or severe skin reaction develop. Liver injury has also been reported. Contact your doctor promptly if liver problems are suspected; treatment must be stopped if liver injury is suspected. Liver monitoring is recommended during HIV-1 treatment and should be considered during HIV-1 prevention use. Depressive disorders have been reported when cabotegravir tablets are used with EDURANT for HIV-1 treatment. Contact your doctor promptly if symptoms of depression develop.

When to call your doctor

- Seek medical attention immediately for a serious allergic reaction or severe skin reaction. - Contact your doctor promptly if you feel depressed or your depression worsens.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.

Before taking this medicine

Cabotegravir tablets (VOCABRIA) must not be taken by anyone who has previously had an allergic reaction to cabotegravir, or alongside carbamazepine, oxcarbazepine, phenobarbital, phenytoin, rifampin or rifapentine. These medicines can lower cabotegravir levels and reduce its effectiveness. CABENUVA must not be used with rifabutin. For HIV prevention (PrEP), VOCABRIA must not be used if HIV-1 status is positive or unknown. PrEP requires comprehensive care to reduce the risk of acquiring HIV-1. Serious allergic reactions, including severe skin reactions such as Stevens–Johnson syndrome and toxic epidermal necrolysis, have been reported. Stop VOCABRIA immediately and seek medical attention if signs of an allergic reaction develop. Liver injury has been reported. Liver blood tests are recommended during HIV treatment; clinical assessment and laboratory monitoring should be considered during PrEP. VOCABRIA should be stopped if liver injury is suspected. Depressive disorders have been reported when VOCABRIA is used with EDURANT for HIV treatment. Depressive symptoms need prompt medical assessment. Interactions

For cabotegravir tablets, medicines that increase the activity of an enzyme called UGT1A1 may lower cabotegravir levels in the blood. Taking other antiretroviral medicines alongside cabotegravir for HIV pre-exposure prophylaxis (PrEP) is not recommended. Ask your doctor to check your medicines for interactions.
