About Venclexta
Overview
Venclexta contains venetoclax, a medicine used to treat adults with chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL). It is also used with azacitidine, decitabine or low-dose cytarabine for newly diagnosed acute myeloid leukemia (AML) in adults aged 75 or older, or adults whose other health conditions rule out intensive induction chemotherapy. It works by blocking BCL-2, a protein that helps cancer cells survive, helping restore the cells’ natural process of self-destruction.
What it treats
- Chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL) in adults. - Newly diagnosed acute myeloid leukemia (AML) in adults aged 75 or older, or adults with other medical conditions that prevent intensive induction chemotherapy. For AML, Venclexta is used with azacitidine, decitabine or low-dose cytarabine.
Safety information
Side effects
Common side effects vary with the condition being treated and the medicines used with Venclexta. For CLL/SLL, when used alone or with obinutuzumab or rituximab, common side effects include low white blood cell, platelet or red blood cell counts, diarrhea, nausea, upper respiratory tract infections, cough, muscle or joint pain, tiredness and swelling. With acalabrutinib, common side effects include low white blood cell counts, headache, diarrhea, muscle or joint pain and COVID-19. For AML, when used with azacitidine, decitabine or cytarabine, common side effects include nausea, diarrhea, constipation, vomiting, low blood cell counts, fever with low white blood cell counts, tiredness, swelling, fever, pneumonia, shortness of breath, bleeding, rash, abdominal pain, sepsis, muscle or joint pain, dizziness, cough, mouth or throat pain and low blood pressure. Serious risks include tumor lysis syndrome, low neutrophil counts (a type of white blood cell), and infections. Some reactions listed as common, including pneumonia, sepsis and bleeding, can also be serious. Contact your doctor promptly if you develop fever, signs of infection, shortness of breath or bleeding. Blood counts and tumor lysis syndrome risk need monitoring. Measures to reduce tumor lysis syndrome risk include adequate hydration and medicines to lower uric acid; higher-risk patients may need intravenous fluids, frequent monitoring and hospitalization.

Before taking this medicine
For patients with chronic lymphocytic leukemia or small lymphocytic lymphoma (CLL/SLL), Venclexta must not be used with medicines that strongly inhibit CYP3A when treatment begins or during the gradual dose-increase phase. This combination increases the risk of tumor lysis syndrome. Venclexta can cause tumor lysis syndrome, low neutrophil counts and infections. Your care team should assess your risk, monitor blood counts and watch for infection. Preventive medicines and adequate hydration are needed; patients at higher risk may need intravenous fluids, closer monitoring or hospitalization. Do not receive live attenuated vaccines before, during or after treatment until your B cells have recovered. Venclexta may harm an unborn baby; effective contraception is advised for patients who can become pregnant. Do not breastfeed during treatment. Severe liver impairment requires a prescriber-directed dose reduction. Venclexta with bortezomib and dexamethasone is not recommended for multiple myeloma outside controlled clinical trials.
Interactions
Tell your doctor if you take strong or moderate CYP3A inhibitors or P-gp inhibitors, as your Venclexta dose may need adjustment. Avoid taking Venclexta with strong or moderate CYP3A inducers. Medicines that are P-gp substrates should be taken at least 6 hours before Venclexta. Ask your doctor whether any of your medicines fall into these groups.

Storage
Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
