Women who use Depo-Provera Contraceptive Injection (Depo-Provera CI) may lose significant bone mineral density. Bone loss is greater with increasing duration of use and may not be completely reversible.
It is unknown if use of Depo-Provera CI during adolescence or early adulthood, a critical period of bone accretion, will reduce peak bone mass and increase the risk for osteoporotic fracture in later life.
Depo-Provera CI is not recommended as a long-term (i.e., longer than 2 years) birth control method unless other options are considered inadequate.
About Depo-Provera
Overview
Depo-Provera is a prescription contraceptive containing medroxyprogesterone acetate, a progestin, supplied in a vial for injection into a muscle. It is used to prevent pregnancy in females who can become pregnant. It works mainly by preventing eggs from maturing and being released, and by thickening cervical mucus. Use for longer than two years is not recommended unless other birth control options are considered inadequate.

Safety information
Side effects
Common side effects include irregular menstrual bleeding or spotting, headache, abdominal pain or discomfort, weight gain, nervousness, dizziness and reduced sex drive. Serious risks include loss of bone mineral density, blood clots, breast cancer, meningioma and severe allergic reactions. Severe allergic reactions require emergency medical treatment. Contact a doctor if you develop jaundice (yellowing of the skin or eyes) or liver problems. If you become pregnant or have severe abdominal pain while using Depo-Provera, contact a doctor because an ectopic pregnancy (pregnancy outside the womb) must be considered. Your doctor should stop Depo-Provera if you develop a blood clot, are diagnosed with meningioma, or develop jaundice or changes in liver function. People with diabetes or a strong family history of breast cancer need careful monitoring.

Before taking this medicine
Do not use Depo-Provera if you have: • Active inflammation of a vein with a blood clot, a current or past blood-clotting disorder, or disease affecting the blood vessels in the brain. • Known or suspected breast cancer. • An allergy to medroxyprogesterone acetate or any other ingredient. • Significant liver disease. • Vaginal bleeding with no diagnosed cause. Careful monitoring is needed if you have a strong family history of breast cancer or diabetes. Patients should also be monitored for signs and symptoms of meningioma. Treatment must be stopped if a blood clot or meningioma is diagnosed, or if jaundice or liver function problems develop. If pregnancy occurs, stop treatment and contact your prescriber. Pregnancy or severe abdominal pain during use requires assessment for a possible ectopic pregnancy. Severe allergic reactions require emergency medical treatment. If you are breastfeeding, discuss use with your prescriber; the medicine has been detected in breast milk. Depo-Provera is not indicated before the first menstrual period.
Interactions
Some medicines and herbal products that increase the activity of certain enzymes, including CYP3A4, may make Depo-Provera less effective at preventing pregnancy. Tell your doctor about any medicines or herbal products you use. If you use an enzyme-inducing product with Depo-Provera, use a backup or alternative method of contraception.
