About Genotropin
Overview
Somatropin is a laboratory-made growth hormone identical to human growth hormone. It treats growth failure in children with growth hormone deficiency, Prader-Willi syndrome, Turner syndrome or idiopathic short stature, and in children born small for gestational age. It also treats growth hormone deficiency in adults, whether it began in childhood or adulthood. It works by activating growth hormone receptors on cells, supporting bone growth and protein production.
What it treats
- Growth failure in children with growth hormone deficiency, Prader-Willi syndrome, Turner syndrome or idiopathic short stature, or who were born small for their gestational age. - Growth hormone deficiency in adults, whether it began in childhood or adulthood.
Safety information
Side effects
Common side effects include headache, injection-site reactions or rashes, and loss of fat under the skin at injection sites. Fluid retention may also occur, especially in adults, causing swelling, joint pain or carpal tunnel syndrome. Seek prompt medical attention for an allergic reaction, as serious reactions can occur. Contact a doctor for persistent severe abdominal pain, which may indicate inflammation of the pancreas; a new limp or hip or knee pain in a child; or signs of upper airway blockage or pauses in breathing during sleep in a child with Prader-Willi syndrome. Other serious risks include increased pressure inside the skull, newly apparent diabetes or impaired blood sugar control, reduced cortisol levels, newly apparent or worsening underactive thyroid, and worsening of an existing spinal curvature. Existing tumours need monitoring for growth or recurrence. Childhood cancer survivors treated with somatropin have an increased risk of a second tumour, particularly meningioma in those who previously received radiation to the head.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
Somatropin should not be used in people with: • Acute critical illness following open-heart or abdominal surgery, multiple accidental injuries, or acute respiratory failure. • Active cancer, or evidence that a brain tumour is growing or has returned. Any previous cancer must be inactive and its treatment completed before starting therapy. • A known allergy to somatropin or any ingredient in the medicine. • Active proliferative or severe non-proliferative diabetic retinopathy, forms of diabetes-related eye disease. It must not be used in children with Prader-Willi syndrome who are severely obese, have a history of upper airway obstruction or sleep apnoea, or have severe breathing impairment. Sudden deaths have been reported in these patients. It should not be used to promote growth in children whose growth plates have closed. Conditions requiring caution include previous tumours, diabetes or impaired glucose tolerance, low adrenal hormone levels, an underactive thyroid and existing scoliosis. Patients with previous tumours need monitoring for growth or recurrence. Childhood cancer survivors treated with somatropin have an increased risk of a second tumour, particularly meningioma after radiation to the head. Blood glucose, thyroid function and adrenal function may need monitoring. Somatropin can cause fluid retention, swelling, joint pain and carpal tunnel syndrome, especially in adults. Raised pressure inside the skull may occur; swelling of the optic nerve must be excluded before treatment. Children with Prader-Willi syndrome need assessment for airway obstruction and sleep apnoea before treatment; treatment should be stopped if these signs develop. A new limp or hip or knee pain in a child needs medical assessment. Persistent severe abdominal pain may indicate pancreatitis. Seek prompt medical attention for an allergic reaction. If acute critical illness develops during treatment, the prescriber must weigh the potential benefits and risks of continuing.
Interactions
Tell your doctor if you take glucocorticoids, oral estrogen, insulin, oral diabetes medicines, or medicines processed by cytochrome P450 liver enzymes. These treatments may need adjustment or closer monitoring when used with somatropin. Somatropin may make it necessary to start glucocorticoid replacement therapy or adjust existing replacement treatment for low adrenal hormone levels. Oral estrogen may affect how much somatropin your prescriber determines you need. Your doctor should manage any treatment changes.

Storage
Store in the original carton to protect it from light. Check the pack for whether it must be refrigerated before first use and how long it may be kept at room temperature afterwards. Keep all medicines out of the sight and reach of children.
