About Orladeyo

Overview

Orladeyo (berotralstat) is a prescription medicine used to help prevent attacks of hereditary angioedema (HAE). It blocks plasma kallikrein, an enzyme involved in producing bradykinin, a substance that causes swelling and pain in HAE. Orladeyo should not be used to treat an HAE attack that has already started.

What it treats

Prevention of hereditary angioedema (HAE) attacks in adults and children aged 2 years and older. Not for treating acute HAE attacks.

Side effects

Common side effects include abdominal pain, vomiting, diarrhea, back pain, and acid reflux (gastroesophageal reflux disease). A serious risk is QTc prolongation, a change in the heart’s electrical activity. Contact your doctor if this is detected. The risk increases at higher-than-recommended doses. Do not take extra doses or more than prescribed.

Before taking this medicine

The label lists no formal contraindications. Orladeyo is not recommended for people with end-stage kidney disease. Avoid use in children aged 2 to under 12 years with severe kidney impairment. Taking more than the recommended amount can increase QTc prolongation, a change in the heart’s electrical activity. Do not take extra doses or more than prescribed.

Interactions

Avoid taking Orladeyo with medicines that increase the activity of P-glycoprotein (P-gp). Orladeyo can also increase levels of certain medicines processed by CYP2D6, CYP3A4 or P-gp. If even a small increase could cause serious side effects, your doctor may need to monitor you closely or adjust the other medicine’s dose. Ask your doctor to check your other medicines for these interactions.

Storage

Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
