Long-term Safety of Topical Calcineurin Inhibitors Has Not
Been Established
malignancy (e.g., skin and lymphoma) have been reported in patients treated with
topical calcineurin inhibitors, including PROTOPIC Ointment.
Therefore:
Continuous long-term use of topical calcineurin inhibitors, including
PROTOPIC Ointment, in any age group should be avoided, and application limited
to areas of involvement with atopic dermatitis.
PROTOPIC Ointment is not indicated for use in children less than 2 years of
age. Only 0.03% PROTOPIC Ointment is indicated for use in children 2-15 years of
age.
About Protopic
Overview
Protopic contains tacrolimus, a medicine used on the skin. It treats moderate to severe atopic dermatitis in adults and children aged 2 years and older who do not have a weakened immune system. It is a second-line option when other prescription skin treatments have not worked well enough or are not advisable. Tacrolimus works by suppressing immune system activity.
What it treats
Moderate to severe atopic dermatitis (eczema) in adults and children aged 2 years and older who are not immunocompromised, when other prescription skin treatments have not worked adequately or are not advisable. Used as second-line treatment for short-term or intermittent longer-term use.
Safety information
Side effects
Commonly reported side effects during treatment with Protopic include skin burning, itching, redness, headache and flu-like symptoms. Other reported effects include skin tingling or increased sensitivity, acne, rash and inflammation around hair follicles. Alcohol intolerance has also been reported. Contact a doctor promptly for an allergic reaction or skin infection, including herpes-related infections. Serious allergic reactions have been reported in clinical studies. Serious problems reported after approval include seizures, severe infections, kidney impairment or acute kidney failure, lymphoma and skin cancers, including melanoma. Seek immediate medical attention for a seizure or severe allergic reaction, and prompt medical advice for suspected serious infection or kidney problems. The frequency of these post-approval events is unknown, and the reports do not establish whether Protopic caused them.
