About Scemblix

Overview

Scemblix (asciminib) is a kinase inhibitor used to treat adults with Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML) in the chronic phase. It is indicated for newly diagnosed or previously treated disease, including disease with the T315I mutation. Approval for newly diagnosed disease is accelerated, based on major molecular response rates, and continued approval may depend on confirmation of clinical benefit in further trials. It works by blocking the activity of BCR::ABL1, an abnormal protein involved in this leukemia.

What it treats

Scemblix is used to treat adults with Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML) in the chronic phase: - Newly diagnosed disease. This use has accelerated approval based on major molecular response rate; continued approval may depend on confirmation of clinical benefit in further trials. - Previously treated disease. - Disease with the T315I mutation.

Safety information

Side effects

Common side effects of Scemblix include muscle and bone pain, rash, tiredness, upper respiratory tract infections, headache, abdominal pain, joint pain and diarrhea. Blood tests may show reduced blood cell counts, hemoglobin or calcium, and increased pancreatic or liver enzymes, cholesterol, triglycerides, uric acid or creatine kinase. Serious side effects that warrant contacting a doctor include severe reductions in platelets or neutrophils (a type of white blood cell), pancreatic problems including pancreatitis, high blood pressure, allergic reactions, and heart or blood vessel problems. Contact your doctor if you develop abdominal symptoms, as these may need assessment for pancreatitis. Regular blood counts, pancreatic enzyme tests and blood pressure checks are needed during treatment. Your doctor may need to interrupt, adjust or stop treatment if serious side effects occur. Scemblix can harm an unborn baby. If you could become pregnant, use effective contraception.

When to call your doctor

• Seek medical attention immediately for abdominal pain if blood tests also show raised lipase (a pancreatic enzyme), as this needs assessment for inflammation of the pancreas.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.

Interactions

Scemblix should not be taken with rosuvastatin or with itraconazole oral solution containing hydroxypropyl-β-cyclodextrin. Tell your doctor if you use either medicine. Other interactions your doctor should check include: • Strong CYP3A4 inhibitors: close monitoring for side effects may be needed, depending on your prescribed Scemblix regimen. • Certain medicines processed by CYP3A4: monitoring or avoiding the combination may be needed, depending on your regimen. • Medicines processed by CYP2C9: these combinations should be avoided. If unavoidable, your prescriber may need to adjust the other medicine or choose an alternative. • Certain medicines transported by P-gp, and medicines transported by BCRP other than rosuvastatin: close monitoring for side effects is needed. Your doctor or pharmacist can check whether your medicines belong to these groups.

Storage

Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
