Hepatotoxicity
Clinically significant and potentially life-threatening liver injury, including acute liver failure requiring transplant, has been reported in patients treated with AUBAGIO in the postmarketing setting. Concomitant use of AUBAGIO with other hepatotoxic drugs may increase the risk of severe liver injury.
Obtain transaminase and bilirubin levels within 6 months before initiation of AUBAGIO therapy. Monitor ALT levels at least monthly for six months after starting AUBAGIO. If drug induced liver injury is suspected, discontinue AUBAGIO and start an accelerated elimination procedure with cholestyramine or charcoal. AUBAGIO is contraindicated in patients with severe hepatic impairment. Patients with pre-existing liver disease may be at increased risk of developing elevated serum transaminases when taking AUBAGIO.
Embryofetal Toxicity
AUBAGIO is contraindicated for use in pregnant women and in females of reproductive potential who are not using effective contraception because of the potential for fetal harm. Teratogenicity and embryolethality occurred in animals at plasma teriflunomide exposures lower than that in humans. Exclude pregnancy before the start of treatment with AUBAGIO in females of reproductive potential. Advise females of reproductive potential to use effective contraception during AUBAGIO treatment and during an accelerated drug elimination procedure after AUBAGIO treatment. Stop AUBAGIO and use an accelerated drug elimination procedure if the patient becomes pregnant.

About Aubagio
Overview
Aubagio is a prescription medicine containing teriflunomide, supplied as film-coated tablets. It treats relapsing forms of multiple sclerosis (MS) in adults, including clinically isolated syndrome, relapsing-remitting disease and active secondary progressive disease. It blocks an enzyme involved in making building blocks for cells and may reduce the number of activated immune cells in the brain and spinal cord, although its exact effect in MS is not fully understood.
What it treats
Relapsing forms of multiple sclerosis (MS) in adults, including: - Clinically isolated syndrome - Relapsing-remitting MS - Active secondary progressive MS
Safety information
Side effects
Common side effects include headache, diarrhea, nausea, hair loss and increased levels of ALT, a liver enzyme measured in blood tests. Serious side effects include liver damage, reduced white blood cell counts and infections, severe allergic reactions, serious skin reactions, skin or mucosal ulcers, impaired wound healing, peripheral nerve damage, increased blood pressure and respiratory problems. Pancreatitis has been reported in pediatric patients. Contact a doctor promptly if you develop signs of infection, skin or mucosal ulcers, wounds that are not healing, or symptoms of nerve damage. Seek urgent medical help for a severe allergic reaction or serious skin reaction. These reactions require stopping Aubagio and treatment to speed its removal from the body. Blood pressure should be checked before and during treatment.

When to call your doctor
If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
Do not take Aubagio if you are pregnant, have severe liver impairment, are allergic to teriflunomide or any ingredient in Aubagio, or are currently taking leflunomide. Do not start treatment if you have an active infection. Aubagio may lower white blood cell counts. A recent complete blood count should be available before treatment starts. Watch for signs of infection and contact your prescriber if they occur; treatment may need to be suspended for a serious infection. Blood pressure should be checked before and during treatment because Aubagio may increase it. Stop taking Aubagio and seek urgent medical help if you develop a severe allergic reaction, swelling of the face or throat, or a severe skin reaction. Your clinician will need to arrange treatment to remove the medicine more quickly from your body. Tell your prescriber if you develop skin or mucous membrane ulcers, wounds that are not healing properly, or numbness, tingling or other symptoms of nerve damage. These may require assessment and stopping treatment.
Interactions
Tell your doctor about all medicines you take, especially warfarin, rosuvastatin and oral contraceptives containing ethinylestradiol or levonorgestrel. Aubagio may lower INR in people taking warfarin, so INR monitoring is needed. It may increase exposure to rosuvastatin and to ethinylestradiol and levonorgestrel. Your doctor should review your rosuvastatin prescription and choose an appropriate oral contraceptive. Aubagio may also increase exposure to medicines affected by CYP2C8, OAT3, BCRP or OATP1B1/B3, and decrease exposure to medicines affected by CYP1A2. Your doctor can check whether your medicines are affected and monitor you as needed.

Storage
