About Ayvakit

Overview

Ayvakit (avapritinib) is a prescription medicine called a kinase inhibitor, supplied as tablets. It treats adults with gastrointestinal stromal tumour (GIST) that cannot be removed by surgery or has spread and has a PDGFRA exon 18 mutation, including D842V. It also treats adults with advanced or indolent systemic mastocytosis, but is not recommended for these conditions when platelet counts are below 50 × 10⁹/L. It works by blocking certain KIT and PDGFRA proteins whose altered activity can contribute to tumour and mast cell growth.

What it treats

- Gastrointestinal stromal tumor (GIST) in adults that cannot be removed by surgery or has spread, and has a PDGFRA exon 18 mutation, including D842V. - Advanced systemic mastocytosis in adults, including aggressive systemic mastocytosis, systemic mastocytosis with an associated blood cancer, and mast cell leukemia. - Indolent systemic mastocytosis in adults. Not recommended for advanced or indolent systemic mastocytosis in patients with platelet counts below 50 × 10⁹/L.

Side effects

Common side effects vary by the condition being treated: • GIST: swelling, nausea, tiredness or weakness, problems with thinking, vomiting, reduced appetite, diarrhea, increased tears, abdominal pain, constipation, rash, dizziness and hair color changes. • AdvSM: swelling, diarrhea, nausea and tiredness or weakness. • ISM: swelling around the eyes, dizziness, swelling in the arms or legs, and flushing. Serious side effects include bleeding inside the skull and changes in thinking or mental function. Seek immediate medical attention if bleeding inside the skull is suspected. If it occurs, Ayvakit must be permanently stopped. Contact your doctor promptly about changes in thinking or mental function; treatment may need to be interrupted, adjusted or stopped. Ayvakit can also cause reactions to sunlight. Limit direct ultraviolet exposure and contact your doctor if a reaction occurs. Ayvakit can harm an unborn baby. Females and males who can have children should use effective contraception.

When to call your doctor

- Seek medical attention immediately for new or worsening difficulty thinking clearly. - Contact your doctor promptly if your skin reacts unusually to sunlight or ultraviolet light.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.

Before taking this medicine

The FDA label lists no contraindications for Ayvakit. However, important warnings and precautions apply: Ayvakit must be permanently stopped if any bleeding inside the skull occurs, regardless of severity. Effects on thinking and memory can occur. Depending on their severity, the prescriber may interrupt, adjust or permanently stop treatment. Ayvakit may cause sensitivity to sunlight. Limit direct exposure to ultraviolet light. Ayvakit can harm an unborn baby. Females and males of reproductive potential should use effective contraception. Do not breastfeed during treatment.

Interactions

Avoid taking Ayvakit with medicines that are strong or moderate CYP3A inhibitors or inducers. Ask your doctor to check whether any of your medicines fall into these groups. If you use a hormonal contraceptive containing ethinyl estradiol, discuss it with your doctor. Recommendations for using it with Ayvakit depend on the Ayvakit dose.

Storage
