About Brukinsa
Overview
Brukinsa (zanubrutinib) is a prescription medicine supplied as capsules for treating certain blood cancers in adults. It is used for Waldenström’s macroglobulinemia, chronic lymphocytic leukemia and small lymphocytic lymphoma. It is also used for mantle cell lymphoma after at least one prior treatment; marginal zone lymphoma that has returned or not responded after at least one anti-CD20 treatment; and, with obinutuzumab, follicular lymphoma that has returned or not responded after two or more systemic treatments. It works by blocking Bruton’s tyrosine kinase (BTK), a protein involved in signals that help B cells multiply and move within the body.
What it treats
Brukinsa is used in adults to treat: - Mantle cell lymphoma (MCL) after at least one previous treatment. - Waldenström’s macroglobulinemia (WM). - Marginal zone lymphoma (MZL) that has returned or not responded to treatment, after at least one anti-CD20-based treatment regimen. - Chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL). - Follicular lymphoma (FL) that has returned or not responded to treatment, together with obinutuzumab, after at least two previous lines of systemic treatment.
Safety information
Side effects
Common side effects of Brukinsa include low neutrophil counts (a type of white blood cell), low platelet counts, upper respiratory tract infections, bleeding, and muscle or bone pain. Serious risks include bleeding, infections (including opportunistic infections), low blood cell counts, new cancers (including skin cancers), abnormal heart rhythms, and liver injury. Contact your doctor if you notice bleeding, signs of infection, or symptoms of an abnormal heart rhythm. Blood counts and liver function need monitoring during treatment. Use sun protection because of the risk of skin cancer. Brukinsa can harm an unborn baby; women who could become pregnant should use effective contraception.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
The FDA label lists no contraindications for Brukinsa. Caution and monitoring are needed for bleeding, infections (including opportunistic infections), low blood cell counts, abnormal heart rhythms and liver injury. Blood counts and liver function should be monitored during treatment. Other cancers, including skin cancers, have occurred; monitoring and sun protection are advised. Brukinsa can harm an unborn baby. Women who could become pregnant should use effective contraception. Do not breastfeed during treatment.
Interactions
Tell your doctor if you take medicines that are moderate or strong CYP3A inhibitors, as your Brukinsa dose may need adjustment. Strong or moderate CYP3A inducers should generally not be taken with Brukinsa. If a moderate CYP3A inducer is used, your prescriber may recommend adjusting your Brukinsa dose.

Storage
