About Calquence

Overview

Calquence contains acalabrutinib, a kinase inhibitor supplied as capsules. It is used to treat adults with chronic lymphocytic leukemia (CLL), small lymphocytic lymphoma (SLL), or mantle cell lymphoma (MCL) after at least one previous treatment. For previously untreated MCL, it is used with bendamustine and rituximab in adults who are not eligible for a stem cell transplant using their own cells. It works by blocking Bruton tyrosine kinase (BTK), a protein involved in signals that help B cells multiply and move.

What it treats

- Previously untreated mantle cell lymphoma (MCL) in adults who are not eligible for an autologous stem cell transplant, used with bendamustine and rituximab. - Mantle cell lymphoma (MCL) in adults who have received at least one previous treatment. - Chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL) in adults.

Safety information

Side effects

Common side effects of Calquence include upper respiratory tract infections, diarrhea, headache, and muscle, bone or joint pain. Serious side effects that require medical attention include serious infections, including infections that occur when immune defenses are weakened; bleeding; low blood cell counts; irregular heart rhythms; and liver injury. Contact your doctor promptly if you develop signs of infection, bleeding or an irregular heartbeat. Other cancers, including skin cancers and other solid tumors, have also occurred. Use sun protection. Blood tests may show reduced neutrophils or lymphocytes (types of white blood cells), reduced platelets, or increased uric acid. Your doctor should monitor your blood counts regularly and your liver function throughout treatment.

- Seek immediate medical attention for signs or symptoms of infection. - Seek immediate medical attention for bleeding. - Seek immediate medical attention for an irregular heartbeat.

If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.

Before taking this medicine

The FDA label lists no formal contraindications for Calquence (acalabrutinib). However, use should be avoided in people with severe liver impairment. Calquence may harm an unborn baby and cause difficult labour. Do not breastfeed during treatment. Important risks requiring caution and monitoring include serious and opportunistic infections, bleeding, low blood cell counts, heart rhythm problems, and liver toxicity, including drug-induced liver injury. Blood counts and liver function should be monitored regularly throughout treatment. Infections should be treated promptly. Other cancers, including skin cancers and other solid tumours, have occurred. Use sun protection.

Interactions

Calquence should not be taken with strong CYP3A inhibitors or strong CYP3A inducers, which affect an enzyme involved in processing medicines. Your prescriber may recommend a dose adjustment for these interactions. Avoid taking Calquence with proton pump inhibitors (PPIs), which reduce stomach acid. Other acid-reducing medicines called H2-receptor antagonists, and antacids, need to be taken at separate times from Calquence. Tell your doctor if you use any of these medicines and ask how to manage them.

Storage

Store below 25°C in a dry place, in the original pack, away from direct light and moisture. Do not keep it in a bathroom. Keep all medicines out of the sight and reach of children.
