About Depacon
Overview
Depacon contains valproate sodium, a prescription medicine given into a vein when taking valproate by mouth is temporarily not possible. It is used alone or with other seizure medicines to treat complex partial seizures and simple or complex absence seizures. It is also used with other seizure medicines for multiple seizure types that include absence seizures. Exactly how it works is not established, but it may increase levels of a chemical messenger in the brain called GABA.

For these conditions, intravenous valproate sodium is used when oral administration of valproate is temporarily not feasible.
Safety information
Side effects
Common side effects reported in studies include dizziness, headache, nausea, sleepiness, tingling, weakness, and pain or irritation at the injection site. Other reported effects include vomiting, abdominal pain, diarrhea, tremor, sweating and changes in taste. Contact a doctor promptly for serious side effects, including liver damage or liver failure, inflammation of the pancreas, unusual bleeding or bruising, and blood cell problems. Raised ammonia levels can affect brain function; unexplained extreme tiredness, vomiting or changes in mental state need prompt medical attention. Other serious risks include an abnormally low body temperature and a severe hypersensitivity reaction called DRESS, which can affect several organs. Older adults may develop excessive sleepiness and need monitoring of food and fluid intake. Exposure during pregnancy can cause birth defects, reduced IQ and neurodevelopmental disorders in the child. Contact your doctor promptly if you are pregnant or planning a pregnancy.

Before taking this medicine
Do not use Depacon if you have liver disease or significant liver dysfunction, an allergy to valproate sodium, a urea cycle disorder, or a known mitochondrial disorder caused by a POLG gene mutation, such as Alpers-Huttenlocher syndrome. It must not be used in children under two years of age with a suspected POLG-related disorder. Valproate must not be used to prevent migraine headaches during pregnancy or in women who could become pregnant and are not using effective contraception. It can cause birth defects, reduced IQ and neurodevelopmental disorders in children exposed during pregnancy. For epilepsy or bipolar disorder, it should not be used during pregnancy, when planning pregnancy, or in women who could become pregnant unless other medicines have failed to control symptoms adequately or are otherwise unacceptable. Important precautions include serious liver injury, pancreatitis, bleeding and other blood disorders, high blood ammonia levels that can affect brain function, abnormally low body temperature, and severe allergic reactions affecting multiple organs (DRESS). Liver tests, platelet counts and blood-clotting tests require monitoring. Unexplained marked sleepiness, vomiting or changes in mental state require assessment for raised ammonia levels. Taking topiramate alongside valproate also requires attention to ammonia levels and low body temperature. Children under two years of age have a considerably higher risk of fatal liver injury. Older adults need monitoring for excessive sleepiness and adequate food and fluid intake.
Interactions
Tell your doctor about all medicines you take, especially: • Phenytoin, carbamazepine, phenobarbital, primidone or rifampin, which can lower valproate levels, and felbamate, which can raise them. Closer monitoring may be needed when these medicines are started or stopped. • Aspirin, carbapenem antibiotics or estrogen-containing hormonal contraceptives. Your doctor may need to monitor valproate levels. • Diazepam, ethosuximide, lamotrigine or phenytoin. Valproate can affect how these medicines are processed or bound to proteins in the blood. • Rufinamide, amitriptyline, nortriptyline, propofol, warfarin or zidovudine. Your doctor may need to adjust treatment when these are used with valproate. • Topiramate. Using it with valproate can cause high ammonia levels in the blood and impaired brain function (encephalopathy).

Storage
Store in the original carton to protect it from light. Check the pack for whether it must be refrigerated before first use. Keep all medicines out of the sight and reach of children.
