About Diprivan
Overview
Diprivan (propofol) is an intravenous medicine used for general anesthesia and sedation. It is used to start general anesthesia in patients aged 3 years and older and maintain it in patients aged 2 months and older. In adults, it is also used for monitored anesthesia care sedation, sedation alongside regional anesthesia, and intensive care sedation in patients who are intubated and mechanically ventilated. Its exact action is not fully understood, but it is thought to enhance the calming effects of a chemical messenger called GABA in the brain.
What it treats
• Starting general anesthesia in patients aged 3 years and older. • Maintaining general anesthesia in patients aged 2 months and older. • Starting and maintaining monitored anesthesia care (MAC) sedation in adults. • Sedation alongside regional anesthesia in adults. • Intensive care unit (ICU) sedation in adults who are intubated and mechanically ventilated.
Safety information
Side effects
Common side effects include a slow, fast or irregular heartbeat; low or high blood pressure; reduced blood flow from the heart; involuntary movements; temporary pauses in breathing; rash or itching; burning, stinging or pain at the injection site; and increased blood fat levels. Acid buildup in the blood due to breathing problems can occur while being taken off a ventilator. Tell the treating doctor immediately about breathing difficulties, pauses in breathing, wheezing, swelling, hives, chest pain, fainting or marked changes in heartbeat. Serious reactions include severe and sometimes fatal allergic reactions, airway blockage, dangerously low blood pressure, heart attack, cardiac arrest and seizures. Children may be particularly susceptible to slowing or stopping of the heartbeat, especially when fentanyl is also given. Rarely, leakage outside the vein can cause pain, swelling, blisters or tissue death. Inflammation or blood clots in a vein have also been reported. Report injection-site symptoms promptly.

Before taking this medicine
Do not use Diprivan if you have a known allergy to propofol or any of its ingredients, or a history of anaphylaxis to eggs, egg products, soybeans or soy products. Serious allergic reactions, sometimes fatal, have been reported. Diprivan can cause a slow heartbeat or cardiac arrest. Children are particularly susceptible to these effects, especially when fentanyl is given at the same time. Strict sterile handling is required to prevent contamination. Diprivan must not be used if contamination is suspected.
Interactions
Opioid pain medicines, such as morphine, meperidine and fentanyl, and sedatives, such as benzodiazepines and barbiturates, may increase Diprivan’s anesthetic or sedative effects. They may also cause greater drops in blood pressure and reduce the amount of blood pumped by the heart. In children, fentanyl given with Diprivan may cause a seriously slow heartbeat. Other medicines used during anesthesia, including nitrous oxide and inhaled anesthetics such as isoflurane, sevoflurane and desflurane, may increase Diprivan’s effects, including effects on the heart and breathing. Tell your doctor if you take valproate. It may increase propofol levels in the blood, requiring close monitoring for increased sedation or reduced heart and breathing function.

Storage
Store in the original carton below 25°C, protected from light. Do not freeze, and use immediately once opened. Keep all medicines out of the sight and reach of children.
