FDA boxed warningRisk of thyroid c-cell tumors
In both male and female rats, tirzepatide causes dose-dependent and treatment-duration-dependent thyroid C-cell tumors at clinically relevant exposures. It is unknown whether MOUNJARO causes thyroid C-cell tumors, including medullary thyroid carcinoma (MTC), in humans as human relevance of tirzepatide-induced rodent thyroid C-cell tumors has not been determined
MOUNJARO is contraindicated in patients with a personal or family history of MTC or in patients with Multiple Endocrine Neoplasia syndrome type 2 (MEN 2)
. Counsel patients regarding the potential risk for MTC with the use of MOUNJARO and inform them of symptoms of thyroid tumors (e.g., a mass in the neck, dysphagia, dyspnea, persistent hoarseness). Routine monitoring of serum calcitonin or using thyroid ultrasound is of uncertain value for early detection of MTC in patients treated with MOUNJARO

About Mounjaro
Overview
Mounjaro contains tirzepatide and is given as an injection under the skin. It is used alongside diet and exercise to improve blood sugar control in adults and children aged 10 years and older with type 2 diabetes. It is also used to reduce the risk of cardiovascular death, non-fatal heart attack or non-fatal stroke in adults with type 2 diabetes who are at high risk of these events. It activates GIP and GLP-1 receptors, helping the body release insulin and reduce glucagon, a hormone that raises blood sugar, in response to blood sugar levels.
What it treats
- To improve blood sugar control alongside diet and exercise in adults and children aged 10 years and older with type 2 diabetes. - To reduce the risk of cardiovascular death, non-fatal heart attack or non-fatal stroke in adults with type 2 diabetes who are at high risk of these events.
Safety information
Side effects
Common side effects of Mounjaro (tirzepatide) include nausea, diarrhea, decreased appetite, vomiting, constipation, indigestion and abdominal pain. Each was reported in at least 5% of treated patients. Serious risks include inflammation of the pancreas (acute pancreatitis), severe allergic reactions, kidney injury due to fluid loss, severe digestive problems, gallbladder disease and worsening diabetes-related eye disease in people who already have it. Low blood sugar, including severe episodes, can occur when Mounjaro is used with insulin or medicines that stimulate insulin release. The label also warns about the risk of thyroid C-cell tumors. Contact a doctor promptly if a serious side effect is suspected. Stop using Mounjaro if pancreatitis or a serious allergic reaction is suspected, and seek medical advice promptly. Serious allergic reactions reported include anaphylaxis and angioedema. Inhalation of stomach contents into the lungs during general anesthesia or deep sedation has been reported in patients receiving GLP-1 receptor agonists. Tell your healthcare providers about Mounjaro use and any planned surgery or procedure.

When to call your doctor
If symptoms are severe or getting worse quickly, seek emergency care rather than waiting.
Before taking this medicine
Do not use Mounjaro if you or anyone in your family has had medullary thyroid carcinoma (MTC), if you have Multiple Endocrine Neoplasia syndrome type 2 (MEN 2), or if you have had a serious allergic reaction to tirzepatide or any other ingredient in Mounjaro. Mounjaro is not recommended for people with severe gastroparesis. Other stomach and bowel reactions can occur and may be severe. Reactions that cause fluid loss can lead to acute kidney injury and may require kidney function monitoring. Stop using Mounjaro if pancreatitis or a serious allergic reaction is suspected. Seek medical advice promptly for a suspected serious allergic reaction. Using Mounjaro with insulin or medicines that stimulate insulin release increases the risk of low blood sugar, including severe episodes. Your prescriber may need to adjust those medicines. If you have a history of diabetic retinopathy, you should be monitored for worsening eye disease. Gallbladder disease can occur; suspected gallstones require assessment and follow-up. Tell your healthcare team about any planned surgery or procedure. Inhalation of stomach contents into the lungs has been reported during general anesthesia or deep sedation in people receiving GLP-1 receptor agonists. Mounjaro may harm an unborn baby, based on animal studies. If you use an oral contraceptive, switch to a non-oral method or add a barrier method for 4 weeks after starting Mounjaro and for 4 weeks after each dose increase. Never share a Mounjaro KwikPen with another person, even if the needle is changed.
Interactions
Mounjaro slows stomach emptying, which may affect how medicines taken by mouth are absorbed. Tell your doctor about any oral medicines you take.
